A cukorrépa szövettenyésztése során a növény ontogenezisének két fontos lépését nem modellezték ezidáig in vitro; a hosszú szártagú virágzati tengely/szár (A) és a raktározó répatest (B) differenciációját.
A. A cukorrépa kétéves növény ami azt jelenti, hogy az első évben raktározószervet és tőlevélrózsát fejleszt, a második évben pedig a rövid szártagú tőlevélrózsa (répafej) közepén egy hosszú szártagú virágzati tengely (tőszár) differenciálódik amely hordozza a reproduktív szerveket. Az in vitro tenyészetekben annyiban hasonló a helyzet, hogy a szövettenyésztett cukorrépa növénykék is tőlevélrózsás felépítésűek, azaz szemmel látható száruk nincsen. A szár internodális szegmentjeinek in vitro megnyújtásával olyan új explantum forrást nyertünk, amelynek segítségével hatékony direkt növényregenerációs módszert lehetett kidolgozni a rekalcitráns cukorrépa esetében. B. A cukorrépa és cékla csiranövények speciális előkezelésével, majd az ezt követő ún. "long-term" indukciójával sikerült - a burgonya minigumó analógiájára - minirépákat indukálni. A. The sugar beet is a biennial plant, which means that in the first year it develops a storage organ and rosette, and in the second year, a long-stemmed floral axis (stem) differentiates in the middle of the short-stemmed rosette (beet head), which carries the reproductive organs. In vitro cultures, the situation is similar in that the tissue-cultured sugar beet plants also have a sepal structure, i.e. they do not have visible stems. By stretching the internodal segments of the stem in vitro, we obtained a new explant source, with the help of which an effective direct plant regeneration method could be developed in the case of recalcitrant sugar beet. On "thin-cell-layer" explants prepared from the epidermal-subepidermal region of the elongated shoots, we regenerated additional buds and leafy shoots from them. We developed a new method for more efficient in vitro rooting of these shoot cultures (Toldi et al. 1996). In addition to conventional micropropagation, this direct plant regeneration method will - we hope - be suitable for the purpose of genetic transformation mediated by Agrobacterium tumefaciens, A. rhizogenesis and/or gene gun.
B. With the special pre-treatment of sugar beet and beetroot seedlings, followed by the so-called with "long-term" induction, it was possible to induce mini-carrots - on the analogy of potato mini-tubers. This result was also the first international publication in the field of in vitro induction of roots modified for storage (Toldi et al. 1993, 1994abc). With histological examinations - by confirming the presence of the polycambial vascular system - we proved that the mini-carrot is anatomically a real carrot body. In addition, according to our measurements, the mini turnip also accumulates sucrose, so it is also a functional storage organ. Based on all of this, the mini-beet offers an opportunity to study the molecular background of carrot body formation, the environmental factors affecting carrot body formation, and the accumulation of sucrose in the storage organ in an in vitro model system. In addition, it is an in vitro inducible organ rich in parenchymatous cells - it can also be used as a potential protoplast source.
